Abstract: In this study, 10 populations of Lycoris aurea Herb. would be compared with leaf, karyotype and SRAP fingerprint, then to be founded the method of identification of these populations. It would be the basis for exploiting and breeding of L. aurea.
First, we founded that had large variation between the leaves of different populations. According to the type of leaf that could be grouped into five categories: (1) The color of Zhongshan Botanical Garden’s leaf was most dark, light and without white powder in the back; (2) The leaves of Jinfoshan, Chongqing and Enshi Hubei were both large and thin, and the reticulate vein was very clear; (3) Jiangxi and Fujian’s leaves were smooth and fleshy; (4) Mingshan Sichuan, Emeishan and Kunming Yunnan, these leaves were leathery and had more white powder under the leaves. (5) The leaf of Sangzhi Hunan was the narrowest one, and the midrib was always purple in the back.
Second, the chromosome numbers and karyotypes of L. aurea had great variation too, according to the number of chromosomes, the 10 populations also could be divided into five categories: (1) 2n = 12 = 10m +2T, Zhongshan botanical garden; (2) 2n = 14 = 8m +4T, Enshi Hubei, Jinfoshan, Chongqing and Laibin Guangxi; (3) 2n = 15 = 7m +8T, Jiangxi and Fujian; (4) 2n = 16 = 6m +10T, Mingshan and Emeishan Sichuan, Kunming Yunnan; (5) 2n = 18 = 4m +11t +3T, it might be a hybrid between L. aurea having 2n=8m+6T and L. radiate that having 2n=22t, this type was reported for the first time in Lycoris, only the population of Sangzhi Hunan had this karyotype. However, the variation was not only in chromosome’s number, because the karyotypes which had the same number of chromosome were also have variation in the morphology and arm ratio, all of these could be used to distinguish between the different populations of L. aurea.
Final, 64 SRAP primer combinations were used to be amplified, and successfully selected 7 combinations which to be constructed the specific band fingerprints; it could distinguish 7 populations from others. And 11 pairs of combinations were selected to build the combination of fingerprints, 7 combinations could distinguish all of those materials, while the other 4 pairs could be combined again to achieve the same purpose too. So the SRAP marker could be used for identifying the different populations of L. aurea material in this experiment.
